TheADAM(ADisintegrinAndMetalloproteaseDomain)familyisauniquegroupofproteinsthathastheABIlitytocleaveorshedextracellularportionsoftransmembraneproteins.Inaddition,ADAMsareinvolvedincell-cellandcell-matrixinteractions(e.g.,fertilization,muscledevelopmentandneurogenesis).
ADAM17(alsoreferredtoasTumorNecrosisFactorConvertase,TACE)isinvolvedintheprocessingoftumornecrosisfactoralpha(TNF-a)atthesurfaceofthecell,andisalsorequiredforsheddingseveralligandsoftheepidermalgrowthfactorreceptorandmanyothermembraneproteins.TheprodomainofcatalyticallyactiveADAMsisthoughttoactasaninhibitoroftheproteaseviaacysteine-switchmechanism.ProdomainremovalisthereforethoughttobeaprerequisiteforADAM17proteaseactivity.
